Postdoctoral Researcher in Chromatin Dynamics at Extreme Desiccation Tolerance
Department of Plant Biology About the Position We are seeking a highly motivated postdoctoral researcher to join our research group at the Swedish University of Agricultural Sciences (SLU) in Uppsala to investigate how plant cells reorganize their cell nuclei and gene regulatory systems to survive extreme environmental conditions. Certain organisms can survive the loss of nearly all cellular water by entering a state of suspended animation, known as anhydrobiosis. Orthodox plant seeds and so-called resurrection plants—plants with extreme desiccation tolerance—are remarkable examples: differentiated vegetative cells can tolerate severe drying and subsequently resume normal growth after rehydration. How a differentiated cell can undergo such an extreme physiological transition while preserving genome integrity, cellular identity, and the ability to reactivate gene expression remains not fully understood. The project will investigate changes in chromatin and transcription in connection with the transition to the desiccated state and recovery from it, using plants with extreme desiccation tolerance and possibly complementary plant systems. We are particularly interested in understanding whether desiccation is accompanied by the establishment of a specialized and reversible chromatin state, how this state contributes to protecting the genome and halting transcription, and how normal organization of the cell nucleus and normal gene expression are restored after rehydration. This position is affiliated with the research group led by Dr Stefanie Rosa at the Department of Plant Biology, Swedish University of Agricultural Sciences (SLU), Uppsala. Our group studies how the organization and dynamics of chromatin affect the regulation and stability of plant genomes (https://www.slu.se/en/profilepages/r/stefanie-rosa/). The project offers great freedom to develop a new research direction at the intersection of chromatin biology, transcription, cell biology, and extreme stress tolerance. The successful candidate will have access to advanced microscopy infrastructure at Uppsala BioCenter and work in a collaborative research environment encompassing plant cell biology, genetics, genomics, and developmental biology. Your Background We are seeking a creative and highly motivated researcher with a PhD in plant biology, molecular biology, cell biology, genetics, genomics, or a related field. Previous experience with molecular biological methods, cell biology, and plant developmental biology is advantageous. Strong ability to communicate orally and in writing in English is required. Key Responsibilities - Generate isogenic leukemia cell line models carrying clinically relevant TP53 alterations using CRISPR/Cas-based genome editing. - Establish models representing distinct allelic states, including wild-type, heterozygous mutation, homozygous mutation, loss of heterozygosity, and TP53 null state. - Define the effect of TP53 genotype on proliferation, competitive fitness, cell cycle control, apoptosis, senescence-like phenotypes, DNA damage response, genomic stability, and stress adaptation. - Assess how TP53 alterations affect sensitivity and resistance to clinically relevant therapies, including chemotherapy, hypomethylating agents, venetoclax-based combinations, DNA-damaging agents, radiation, and novel targeted treatments. - Contribute to method development for applying Cell Painting to suspension cells and to machine learning models for classification of TP53 variants. Scope of Work Approximately 70–80% experimental/wet lab work, 20–30% data analysis, modeling, and manuscript preparation; the exact balance can be adjusted to the candidate's profile and project needs. Supervision The postdoctoral researcher will be scientifically supervised by the PI(s) of the leukemia/TP53 program and will collaborate closely with senior bioinformaticians, clinicians, and molecular geneticists; they may co-supervise master's/doctoral students in daily laboratory and analysis work. Specific Responsibilities/Authority - Lead planning of experiments and analyses within the TP53 work package, including selection of variants, cell models, and compound conditions. - Make independent decisions on protocol optimization, feature selection, and computational workflows, in consultation with the project leader. - Ensure data quality control, documentation, and reproducibility, and take a leading role in developing manuscripts, presentations, and method descriptions for this project. The overall goal of this project is to deepen fundamental understanding of defect-related physics and provide new insights into the hotly debated topic of 2D material defect passivation, by for the first time utilizing advanced vibrational spectroscopies, time-resolved optical spectroscopy, and microscopy. Responsibilities Being a doctoral student means independently driving projects within a specific doctoral research subject. You collaborate continuously with other doctoral students and researchers to jointly produce results within various projects and have one or more supervisors to support you during your doctoral studies. However, you work largely independently, for example, through work in a laboratory environment and writing articles for journals and results for your doctoral thesis. PhD Student in Evolutionary and Ecological Genetics
Do you want to conduct groundbreaking research in a leading environment within evolutionary biology? The Department of Ecology and Genetics (IEG) at Uppsala University conducts world-leading research in biology, from molecules to ecosystems and global biodiversity patterns. We are now seeking a PhD student who wants to develop their research career with us. IEG at the Evolutionary Biology Centre is an international research environment with access to advanced research infrastructure, such as supercomputing resources (NAISS), SciLifeLab platforms, climate laboratory, greenhouses, and modern laboratories. PhD students can also join the department's graduate school, which offers excellent networking opportunities. The position is offered within the research program for Zoo Ecology. Research includes population and conservation genomics, quantitative genetics, evolutionary ecology, behavioral ecology, and macroecology. Project Description: A central question in evolutionary biology is whether evolutionary changes are repeatable and whether the same genes contribute to repeated phenotypic adaptations. This question has important implications for our ability to predict genetic responses to changing environments. This project aims to integrate mechanistic insights about phenotypic adaptation with genomic data to understand how species develop adaptations to changes in their living environment. The project will utilize data from a large-scale Evolve-and-Resequence experiment on a widespread pest insect, the seed beetle Callosobruchus maculatus. This unique system enables studies of the role of determinism and chance in local adaptation and the evolution of a species' ecological niche. Phenotypic data, RNA-seq, and DNA pool-seq data are available from multiple time points in the ongoing experiment. The Berger group also maintains several other geographic populations of C. maculatus as well as closely related seed beetle species. This provides opportunities for comparative genomics and additional experiments and data collection. Main Duties: The PhD project will involve a combination of experimental laboratory work and bioinformatic analysis, with emphasis on the latter. The exact distribution between tasks may depend on the candidate's interests and project development. PhD students at the Department of Ecology and Genetics (IEG) have the opportunity to participate in teaching and develop their pedagogical CV by taking courses. The type and extent of teaching depends on availability and the student's interests but may not exceed 20% of full-time. Employment and funding are extended corresponding to the time spent on teaching. The PhD student will work with the main supervisor David Berger and co-supervisor, SciLifeLab researcher Dr Gabriela Montejo-Kovacevich, both at IEG. Qualification Requirements: To meet the eligibility requirements for doctoral studies, you must: have a master's degree (second cycle) in evolutionary biology, ecology, bioinformatics, or a related field, or have completed at least 240 higher education credits, of which at least 60 credits at advanced level including an independent project worth at least 15 credits, or have acquired equivalent knowledge in another way (e.g., extensive research experience). Candidates who have not yet completed their degree but are expected to meet the requirements by the start date may apply for the position. The ideal candidate should also: Have a good understanding of central concepts in ecology and/or evolutionary biology/genetics. Have programming experience. Be motivated and curious and enjoy a collaborative environment. Be able to take responsibility for carrying out and completing independent tasks and projects. Be able to express yourself well in both spoken and written English. Meritorious: Experience with statistical analyses applied to quantitative genetic or population genomic data.
