PhD Student in Biology, Environmental Science Focus
Department of Aquatic Resources Doctoral Field: Biology. Assessment and prioritization of marine and coastal restoration measures. There is an acute need for restoration in coastal and marine areas. The restoration challenge is complex, as many areas are affected by large-scale pressures that interact with local effects. To meet this challenge, we need to understand how ecological knowledge can be combined with societal processes, both at local and larger spatial scales. The doctoral position is part of a project that will support effective strategic planning for marine coastal restoration by developing key performance indicators (KPIs) to assess and evaluate the effectiveness of coastal restoration measures based on ecological, social, and economic sustainability aspects. The project group is interdisciplinary and includes researchers in ecology, sociology, and national economics. The project's results will form the basis for restoration measures at both local and national levels. Duties: The main task is to conduct doctoral research under supervision, which includes developing the doctoral student's methodological, theoretical, analytical, written, and oral presentation skills. The doctoral student will use a range of different methods, including stakeholder dialogues, geographic data analysis, and validation and testing of indicators in case studies. You will develop maps of natural values that contribute important knowledge for sustainable coastal planning and identify hotspots where restoration measures would provide the greatest cost-effectiveness in relation to ecological, social, and economic goals. Your work will contribute to increased knowledge about how spatially targeted measures and spatial optimization can affect the effectiveness of restoration efforts, as well as processes for integrating knowledge from multiple sources in management work. You will have close contact with officials from municipalities, regional authorities, and the Swedish Agency for Marine and Water Management. Uppsala University, Department of Medical Sciences Description of the Subject Area Hematology is a central clinical and academic subject area with strong connections to both research and education as well as to highly specialized care. You will be part of Uppsala University's Molecular Precision Medicine research group, where research focuses on both the development of new genomics techniques and their application to clinically relevant questions in pediatric oncology. Research Assistant in In-Cell NMR of RNA Dynamics
The Department of Medical Biochemistry and Microbiology (IMBIM) at Uppsala University is a broad international environment for research and teaching. Project Description The project aims to understand how RNA structure and dynamics contribute to gene regulation in the cellular environment. A particular focus is on developing and applying in-cell NMR to quantify RNA conformational dynamics, including exchange between ground and excited states, directly in living human cells. Using enzymatic circularization of RNA to counteract intracellular degradation, and optimized relaxation-based data acquisition (selective excitation, SOFAST, and R1ρ relaxation dispersion), the project will map how the cellular environment shapes the structural ensemble of biologically relevant RNA molecules. Duties The work is primarily experimental (laboratory and NMR work) and includes: Preparation and purification of isotope-labeled RNA and DNA samples for NMR (in vitro transcription, large-scale production, and enzymatic circularization of RNA hairpins) Introduction of nucleic acids into human cell lines (e.g., electroporation) and preparation of in-cell NMR samples, including controls for cell viability and leakage (supernatant control) Collection and optimization of solution NMR experiments for nucleic acids, including imino proton detection with selective excitation/SOFAST and measurement of longitudinal relaxation (selT1) Measurement of RNA dynamics using 1H and 15N R1ρ relaxation dispersion to quantify populations, exchange rates, and chemical shifts of excited states, in vitro and in-cell Quantification of intracellular nucleic acid concentration and subcellular localization Processing, curve fitting, and analysis of relaxation and dispersion data Documentation and presentation of results Work is conducted in close collaboration with experimental and computationally-oriented researchers in the group.
