Responsibilities The successful candidate will participate in an experimental research project investigating genes involved in susceptibility to liver fibrosis and cirrhosis. The responsibilities include establishment and maintenance of spheroid and organoid cultures from hepatocyte cell lines and primary human hepatocytes as well as generation and maintenance of stable mammalian cell lines. The work also encompasses the use of molecular biological methods, including DNA and RNA isolation, DNA cloning, transfection, PCR, protein expression analyses, and RNA expression profiling. Further, the candidate will perform biochemical analyses such as immunoblotting (Western blot), immunoprecipitation, subcellular fractionation, and siRNA-mediated gene silencing. The project includes metabolic flux experiments with radioactive tracers, as well as immunocytochemistry and in situ hybridization experiments. Analysis of genetic and genomic datasets will be conducted using R- and Linux-based computational tools. The work also includes documentation, analysis, and presentation of experimental results as well as active participation in regular laboratory meetings and scientific discussions. Qualifications The applicant should have a background in biomedical sciences, molecular biology, biotechnology, cell biology, or another relevant life science discipline. Prior experience with organoid or spheroid culture or alternatively liver biology is required, as is practical experience with molecular biological work. Proficiency in spoken and written English is also necessary for the position. It is advantageous to have experience with omics analysis as well as work with mammalian cell culture, including spheroids and/or organoids, and generation of stable cell lines.
