Project Description Our research combines experimental approaches with analyses of human biobank material and clinical databases to understand how mechanical forces regulate gene expression. We have recently mapped transcriptomic changes in arteries from aging humans and rodents (https://doi.org/10.1016/j.jbc.2025.108400; https://doi.org/10.1161/HYPERTENSIONAHA.125.26119) and now aim to clarify the underlying mechanisms by developing transgenic models and investigating their significance for vascular function and vascular disease. A central goal is to model increased expression of the complement system in aging arteries, where the postdoctoral fellow is expected to contribute to collaborative projects with the University of Copenhagen and the University of Gothenburg. To investigate this, you will generate cell-specific and inducible knockout mice by crossing endothelial (Tie1) and smooth muscle cell-specific (Itga8) Cre lines with mice carrying floxed Cfh alleles (https://mice.jax.org/). Genotyping is performed externally, but you will be responsible for colony and tissue sample management, including breeding strategy, cage planning, and tamoxifen treatment for inducible knockout models. You will also plan and conduct experiments in close collaboration with colleagues, which on occasion may involve work outside regular working hours. Vascular phenotypes will be analyzed using histological and physiological methods, including disease models based on hypercholesterolemia and hypertension.
