Subject Microbiology Subject Description The gut microbiota contains approximately 1,000 times more genes than the human genome and constitutes a complex ecosystem that affects the host's physiology through the production of bioactive metabolites. Despite major advances in microbiome research, it remains largely unknown how individual bacterial species and genetic variation between different bacterial strains contribute to microbiota function and the development of metabolic diseases. This project aims to identify and characterize bacterial species and strains within the families Lachnospiraceae and Oscillospiraceae, with particular focus on the genus Faecalibacterium, one of the most prevalent and functionally important bacterial groups in the healthy human gut microbiota. Duties The successful candidate will investigate the functional diversity of Faecalibacterium and its interactions with other gut bacteria through an integrated experimental and bioinformatic approach. The project combines comparative genomic and bioinformatic analyses to examine how genetic variation at the strain level affects bacterial metabolism and functional properties with physiological characterization of bacterial growth, substrate utilization, and production of bioactive metabolites under defined anaerobic culture conditions. Based on our observations that the presence of Faecalibacterium is associated with increased stability in the intestinal microbial community, likely through interactions with bacteria within the families Lachnospiraceae and Oscillospiraceae, the candidate will also isolate and characterize new bacterial strains from fecal samples from healthy donors. These isolates will be used in experimental models to study microbial interactions, cross-feeding, and their significance for host metabolism. The project integrates clinical cohorts, large-scale genomic analyses, anaerobic bacterial culture, metabolomics, and experimental model systems to identify microbial mechanisms and bioactive metabolites that contribute to metabolic health and that may form the basis for future microbiome-based treatments of cardiometabolic diseases. Qualifications Eligible for employment as a postdoctoral researcher is a person who holds a doctoral degree in a relevant subject area, or a foreign degree deemed equivalent to a doctoral degree. This qualification requirement must be met at the latest when the employment decision is made. First priority should be given to applicants who completed their degree at most three years before the application deadline. Those who completed their degree earlier should also be considered if there are special reasons. Furthermore, experience with programming and statistical data analysis in R is required, along with very good ability to express oneself verbally and in writing in English. Experience with isolation, cultivation, and physiological characterization of human gut commensals belonging to the families Lachnospiraceae and Oscillospiraceae, particularly butyrate-producing and/or fiber-degrading bacteria, is highly meritorious. Experience working with fecal cultures from human samples, including selective or enrichment-based isolation strategies under strictly anaerobic conditions, is also meritorious. Additional merit will be given to experience studying bacterial metabolism of dietary components, including dietary fibers, complex carbohydrates, or other diet-related substrates. The unit will be built with four mobile interprofessional teams consisting of physicians, psychologists, social workers, nurses, occupational therapists, and healthcare administrators. In total, the operation will comprise approximately 25 employees. The target group is children and young people with extensive psychiatric and social comorbidity. Many have experience with care breakdowns, repeated placements, and serious mental health issues, such as self-harm, trauma, or behavioral disorders. The catchment area is the entire Västra Götaland region. Initially, the operation will focus on adolescents placed in SiS facilities in the region. Long-term, children and young people placed in HVB facilities and within the Prison Service will also be included. Organizationally, the unit belongs to the Neurology and Child Psychiatry operation at Sahlgrenska University Hospital, within the Child Psychiatric 24-hour Care and Specialist Clinics section. The operation is mobile and meets the young person where they are located. Work is conducted as teamwork with close collaboration between professionals. The Department of Medicine, Division of Molecular and Clinical Medicine, conducts translational research primarily focused on metabolism, diabetes, obesity, and cardiovascular disease with the aim of improving health and healthcare in society. The overarching goal is to create an excellent translational research environment of very high international standard. The Romeo laboratory studies the biological and genetic mechanisms behind metabolic dysfunction-associated steatotic liver disease (MASLD) and its progression to liver fibrosis and cirrhosis. Our research combines human genetics with experimental models to identify disease mechanisms and therapeutic targets. We use state-of-the-art cell culture systems, liver organoids, and molecular biological methods to investigate signaling pathways involved in liver injury and fibrosis. Responsibilities The successful candidate will participate in an experimental research project investigating genes involved in susceptibility to liver fibrosis and cirrhosis. The responsibilities include establishment and maintenance of spheroid and organoid cultures from hepatocyte cell lines and primary human hepatocytes as well as generation and maintenance of stable mammalian cell lines. The work also encompasses the use of molecular biological methods, including DNA and RNA isolation, DNA cloning, transfection, PCR, protein expression analyses, and RNA expression profiling. Further, the candidate will perform biochemical analyses such as immunoblotting (Western blot), immunoprecipitation, subcellular fractionation, and siRNA-mediated gene silencing. The project includes metabolic flux experiments with radioactive tracers, as well as immunocytochemistry and in situ hybridization experiments. Analysis of genetic and genomic datasets will be conducted using R- and Linux-based computational tools. The work also includes documentation, analysis, and presentation of experimental results as well as active participation in regular laboratory meetings and scientific discussions. The position offers an opportunity to gain practical experience in translational liver research and experimental disease modeling. Qualifications The applicant should have a background in biomedical sciences, molecular biology, biotechnology, cell biology, or another relevant life science discipline. Prior experience with organoid or spheroid culture or alternatively liver biology is required, as is practical experience with molecular biological work. Proficiency in spoken and written English is also necessary for the position. It is advantageous to have experience with omics analysis as well as work with mammalian cell culture, including spheroids and/or organoids, and generation of stable cell lines. Experience with molecular biological techniques such as DNA and RNA extraction, cloning, PCR, transfection, and gene expression analysis is also meriting. Further, experience with biochemical methods, for example Western blotting, immunoprecipitation, and immunocytochemistry, is considered an advantage.
